Usage | Dissolve type I collagen at a concentration of 4 mg/mL in an acidic buffer solution, such as 0.01–0.05 M acetic acid at pH 3.0–3.3 or 0.15 M citrate buffer at pH 3.6, and mix overnight at 4°C. Due to the rigidity and long triple helix conformation of collagen molecules, it is recommended to keep the collagen solution concentration below 4 mg/mL; concentrations above 4 mg/mL will result in high viscosity and difficulty in handling. To dissolve collagen in a neutral buffer, dissolve collagen in 0.01–0.05 M acetic acid, add two volumes of neutral buffer, and then dilute with one volume of buffer. The collagen solution must be kept on ice. Do not leave the neutral collagen solution at room temperature, as collagen will form a gel. |
