Description

Ficin is a cysteine protease isolated from FIG latex (EC 3.4.22.3) with endopeptidase activity for cutting immunoglobulin G molecules in the hinge. Ficin is commonly used to cut mouse IgG1, as this is difficult to cut with papain and pepsin. In the presence of 1mM or 10mM cysteine, ficin produces F(ab ')2 and Fab fragments, respectively. Fab and F(ab ')2 fragments can be isolated from whole IgG and Fc using immobilized protein A or ion exchange chromatography.
