Papain is a cysteine protease (EC 3.4.22.2) with endopeptidase activity for cutting immunoglobulin G molecules in the hinge region. 3 ~50kDa fragments were produced by pyrolysis. Two Fab domains and one Fc domain. Papain digested antibodies do not promote agglutination, precipitation, conditioning, and cleavage. Fab and Fc fragments can be further separated by immobilized protein A or ion exchange chromatography.
