The purification of plant-derived papain was prepared by freeze-drying from crystalline suspension. For best results, the enzyme should be incubated for 30 minutes in a solution containing 1.1 mM EDTA, 0.067 mM mercaptoethanol, and 5.5 mM cysteine hcl. Enzyme dissolved after filtration 0.22 micron, strongly recommended before use. Papain is activated by cysteine, sulfides, and sulfites, and stabilizers include EDTA, cysteine, and dimercaptoethanol.
